1) CELLnTEC – Company Overview CELLnTEC Advanced Cell Systems is a Swiss company specialized in epithelial in vitro systems. They sell a range of media and progenitor cell lines for epithelial cell culture. Chemicon is their worldwide distributor. CELLnTEC’s major products are culture media for epithelial tissues such as skin, airway, prostate, cornea and bladder. 2) Product Line Overview CELLnTEC offers a range of media, specifically for epithelial cell culture. They offer 3 types of media for the culture of a variety of epithelial cell types: a) PCT (progenitor cell targeted), b) complete (defined, serum free) and c) non-defined media. CELLnTEC Media are available for the following epithelial tissues: Epidermis, Airway, Bladder, Prostate, Vaginal, Mammary, Oral, Corneal, Dermis. Most are available in some (or all) of the following 3 different formulations: 1) PCT Media: These are specifically designed to isolate and grow progenitor cells (i.e. adult stem cells) in culture. These media are fully defined, serum, BPE-free, and are particularly effective in increasing the number of progenitor cells in culture and delaying the onset of differentiation. These media also provide extended lifespan of cultures, and better cell morphology. A defined, serum/BPE-free formulation means no need for feeder cells or conditioned media, and eliminates the variables associated with a non-defined formulation. Note: researchers wishing to differentiate their cells will need to switch to a different, non-PCT media after isolation of progenitor cells (Complete or Non-defined) since PCT media keep progenitor cells in their non-differentiated state.Benefits of PCT Media The PCT media are unique in that they target progenitor cells (adult stem cells). In the body, progenitor cells drive growth and regeneration of all tissues throughout the life of the organism. In addition, they have an indefinite life span and are not tumorigenic – the optimum features of a cell culture. To maintain these specific cells in vitro requires a very precise environment. By targeting these cells, CELLnTEC products enable significant improvements over existing alternatives. 5) Media Product configuration Each CELLnTEC media kit consists of 2 parts: Basal Media, 500mls (CnT-XX.BM), stored at 4ºC, 12 month shelf life) Supplements (CnT-XX.S) store at -20ºC, 12 month shelf life) 6) Customers CELLnTEC PCT media mimic the epithelial adult stem cell niche environment in vitro. Any researcher using epithelium cells as a model system is a candidate for CELLnTEC media. Customers are generally of three different types: Experts, General Media Users or Novices. • Experts: These researchers are considered the “Thought Leaders” in their particular field. They generally have their own medium formulation that they have used for a number of years to conduct their research. This customer can be very hard to convert to a new medium but if they do agree to test and use the CELLnTEC medium, others will follow their lead. • General Medium Users: These researchers are knowledgeable of the difficulty to isolate progenitor cells from tissues. They use commercially available medium but most are open to trying a new formulation. Improved isolation efficiency is their primary motivation for trying a new formulation. • Novices: These potential customers have generally stayed away from the isolation and cultivation of cells from tissue due to the low success rate. With CELLnTEC medium’s high isolation efficiency, this previously difficult method is more achievable by the novice to this technique. Researchers not previously isolating cells from tissue are a vastly untapped market. Media can be sold to a variety of different customer profiles. • Dermatology/Skin Cancer • Cancer Biology/Oncology • Urology/Prostate Cancer • Respiratory/Pulmonary • Medicine/Pathology • Regenerative Medicine/Cell Therapy/Tissue Engineering • Ophthalmology/Corneal Research 7) Medium Sampling Program We have found that offering a free sample to test in a side by side comparison with the customer’s current medium is a highly effective way to introduce the CELLnTEC medium into a laboratory. Samples are available for ALL PCT media, and most non-PCT media. (Exceptions are CnT-30, 32, 33, 34, 35, 36 and 39). The product code is CnT-XXsample. (XX reflects media # to be sampled. i.e CnT-30sample) Sample Configuration: The CELLnTEC medium samples are provided as a 100 ml sample size, completely supplemented and ready to use. No additional supplementation is required. The medium has a shelf life of at least six months from the date on the bottle but we ask the customer to commit to test the sample as soon as possible after it is delivered to their laboratory. (The date on the label is the date the supplement has been added to the basal medium.) Important information for the person sampling: • Please emphasize they should refer to the “sampling recommendations protocol” so they get the best possible results (sent with product, also available under web for each catalog #). • The CELLnTEC defined media samples are provided as 100ml samples, completely supplemented, and are ready to use – there is no need for serum or additional components. (The purchased product is supplied as a kit consisting of 500 ml basal medium with separate supplements to be combined before initial use.) • Also, for the majority of isolations, there is no need to use feeder cells, conditioned media or coating matrix when using these media. Customers who are currently using these techniques should test the media with and without them. • When stored at 4ºC, CELLnTEC media have a shelf-life of three months after the date they are received. Customers will get optimal results when their evaluation is within this time frame. • If they are using a trypsinization procedure, bear in mind that serum free media are low in protein and thus will not stop the trypsinization step efficiently. Either add serum to the media for this step or refer to CELLnTEC’s MPEK passage and freezing protocol where we suggest using rProtease. ******* Remember - Follow-up is the most important aspect of a sampling program! ********** Medium Sampling Program - Troubleshooting: Rarely do we hear “It did not work well” because the side by side comparison is what sells our medium. But occasionally it happens – the best response is to ask questions to try to understand the situation. The defined media are developed to perform better than non-defined, FBS and/or BPE containing media; but with the large variation in “homegrown” formulations we can not predict how they will perform in every situation. Below are several replies we have received and our response: My previously isolated cells, growing in my current medium did not grow very well when switched to the CELLnTEC medium. Response: Many times, when switching to a different medium, there is a need to wean your cells off the old medium. This is accomplished by serial dilution of the current medium with the CELLnTEC medium over a period of a week or two. My isolation efficiency was very low compared to my current medium. Response: -What is your current medium? Does it contain FBS of BPE? Did you use trypsin in your isolation protocol. Low isolation efficiency could be due to over trypsinization that was previously stopped by the high protein concentration in your medium. Refer to “Resources” section of the CELLnTEC web site: www.cellntec.com where we recommend the use of rProtease. - What species, strain (if mouse or rat) and what is the age of the donor? CELLnTEC has seen large variation in isolation efficiencies depending on these variables. -Was the CELLnTEC medium tested in side by side comparison to your current medium? How old was the CELLnTEC medium compared to your current medium? I have received the sample but have not/ will not be doing those experiments. – Response: Is there another person in your laboratory (or Department) that is performing isolations or growing progenitor cells that might consider testing the sample? May we have the sample back, so another customer can benefit?
